We have our AD/ADFS hosted in azure on a vm ( not Azure AD) and are using it as identity provider for external applications ,now we have a requirement to enable MFA for a particular SaaS application , is it possible to enable MFA without Azure AD?
Sure, but you will have to use leverage some MFA solution. Its not built-in to ADFS:

Just a follow up question. Is Azure MFA dependent on the third-party application supporting MFA or regardless we can enable MFA ? so for e.g If I have a SaaS application called abc.com and abc.com doesn't support MFA , so for my corporate users to login into abc.com I would like to enable MFA using my ADFS , is that possible given third party application doesn't support MFA?
The MFA is handled by ADFS and however its integrated with ADFS. The 3rd party app itself doesn't know about MFA. As long the as the app supports SAML/Claims etc... thats all matters. In other words, if it works now with ADFS, it will work with ADFS MFA integration as well.
